I have two hard drives, and my main (C:\) is nearly full. If I move the 'Maxis' folder containing my original Sims & the expansions to my other hard drive, will the game still work?

Yes ... it will still work until you re-install your (c:\) with a new windows.
If that happen, you need to re-install the game from it's original version to whatever expansions you might have ... or it won't recognize your 'old' files.
